【摘要】:Thisguideislimitedtoproceduresusedsolelyforthetestingofsubstancestodeterminetheirmutagenicityanddoesnotapplytoothermethodsandusessuchasexploringmechanismsofmutation.Recentevidencesuggeststhatthisassaymeasuresadualgeneticendpoint;therefore,somediscussionoftherelationshipsbetweenmammaliancellmutagenicitytestingresultsandtheresultsobservedbothinpuregenemutationalassaysandincytogeneticassaysisnecessary.However,itisnottheintentofthisguidetodiscussotherrelationshipsbetweenthismammaliancellmutagenicitytestingresultsandtheresultsobservedinothertestsformutagenicityandcarcinogenicity.1.1ThepurposeandscopeofthisguideistopresentbackgroundmaterialandtoestablishcriteriabywhichprotocolsandproceduresforconductingtheL5178Y/TK+/-3.7.2Cmouselymphomamutagenicityassay(commonlyreferredtoasthemouselymphomaassay,(MLA))canbeproperlyunderstoodandevaluated.Thisguideisalsointendedtoaidresearchersandotherstogainabetterunderstandingofthecriticalelementsinvolvedwithmammaliancellmutagenicitytesting.Morespecifically,thisguideisintendedtoprovideforresearcherstheaccomplishmentofthefollowinggoals:1.1.1Provideanunderstandingofthecriticalprocedures(steps)intheperformanceofthismammaliancellmutagenicitytest.1.1.2Providegeneralizedcriteriabywhichresearcherscanevaluateiftheyareproperlyperforming,utilizing,andinterpretingthisassay.1.1.3ProvidecriteriabywhichindividualsresponsibleforevaluatingMLAdatacandetermineiftheexperimentshavebeenproperlyperformedandinterpreted.1.1.4Provideabasisfromwhichnewproceduresanddevelopmentsintestingprocedurescanbeevaluated.1.1.5Provideanunderstandingofthetypesofgeneticdamage(thatis,geneandchromosomemutation)thatmaybedetectedinthismammaliancellmutagenicitytest.

【摘要】:TheserequirementsareapplicabletoIAQGglobalsectorswhenutilizingAerospaceIndustryControlledOtherParty(ICOP)schemesfortheassessmentandcertificationofsupplierqualitysystemsinaccordancewiththerequirementscontainedinEN9104.IAQGmembercompanieshavecommittedtoperformsufficientoversightoftheAerospaceaccreditation/certificationassessmentprocessesinordertoensureconformancetoestablishedrequirementscontainedinEN9104.NOTE:TheIAQGOPMToversightstructureisdepictedgraphicallyinAnnexA.Thepurposeofthisstandardistodocumenttheprocessforoversightofthe9100-seriescertificationofAerospaceQualityManagementSystems(AQMS)program.ThisprocessshallbeimplementedbyparticipatingIAQGmembercompanies,ABs,CBs,AABs,TPABsandtrainingproviderstoensureconsistentconformancewiththerequirementsofEN9104.
